Using normal competent cells in electroporation method? - (May/09/2012 )
Is it possible to electroporate normal competent cells (e.g. JM109) or electrocompetent cells are the only way to go?

You can easily make electrocompetent cells from normal (non-competent) cells. Just grow a culture to OD 0.6, wash twice with cold pure water, and concentrate about 200x (If you start with 200 ml, your final volume should be 1 ml). If you want to freeze them, do a final wash in 10% glycerol.
