immunostaining of cells in culture - (May/04/2012 )

Hi,

I have to test for the presence of stem cells in my cell line by immunohistochemical staining (IHC). I've done IHC on formalin fixedparaffin embedded block but I don't know how I can fiz the cells in culture.

Any advice would be much appreciated.

Culture your cells on cover slips. In the well: wash with PBS, fix with 4%PFA in PBS 5min @RT. Wash with PBS +0.1% Triton X100 2x5min. Block, stain, wash, stain, wash, mount.
