NZY+Broth can be replaced by LB Broth in mutagenesis exp - (May/03/2012 )

Hello everyone,

I am usnig Stratagene Quick change II mutagenesis kit to insert a restriction site in my sequence.
I don't have NZY+ Broth, can I use LB Broth ?
Composition of LB broth (10 g tryptone, 5 g yeast extract, and 10 g NaCl in 1 litre of water)
Compositon of NZY+ broth (10 g of NZ amine (casein hydrolysate), 5 g of yeast extract, 5 g of NaCl in 1 litre of water, before use add supplements; 12.5 ml of 1M MgCl2, 12.5 ml of 1M MgSO4, 20ml of 20% glucose)
1. As I don't have NZY+ Broth, if I add supplements of NZY+ broth to LB broth, will it work ??
2. Or I can replace NZY+ broth with LB Broth ?

Either should work fine. Your problem will not be growth of the cells.

Use of a richer media will give you more transformants, which is helpful but not absolutely necessary. I use SOC media.
