LC3 Autophagy Huh7 - (Apr/03/2012 )

I am looking at autophagy levels using polyclonal LC3 antibody in Huh7 cells. My expectation is that little/no autophagy is occuring in Huh7 cells. However, when I performed a western blot I had very dense LC3I and LC3II bands indicitive of autophagy. Why is a Huh7 cell line (that appears healthy in culture) undergoing autophagy? I am going to treat with an autophagy inhibitor to determine if I can decrease the level of LC3II, but does anybody have an explanation as why I see high levels of both LC3I and LC3II?

Anybody? We use 10% serum, seems to be the standard, autophagy can be induced when serum is low
