i have a very simple question , i cheked my phosphatase labelled secondary body.
I usually diluted 100 ng / ml concentration , from this diluted sample i took little amount and add into a separate tube then add little amount of substrate and it gives me a colour ??
Is this contamination ??

no, your conjugate SHOULD give you color when substrate is added...alll your other reagents should NOT.
Have your "trainer" do the test and watch him/her OR HAVE HIM/HER WATCH YOU.
