Differences between J774.1 and RAW 264.7 murine macrophages? - (Feb/01/2012 )

Hi all, i would like to carry out an investigation on host-pathogen interaction. Just come across there are two types of murine macrophages that been widely used in h-p interaction study. Just wondering, anyone can list out the significant differences among this two cell lines of macrophages? Just know that RAW 264.7 is more capable in producing NOS, but how about J774.1?

Our group have used both RAW and J774 cells and have published on both. The research has been mainly on inducible nitric oxide synthase (iNOS) but we have also looked at Salmonella interactions with J774 cells.
We used J774 cells to produce iNOS for drug screening at the pharmaceutical company that I worked for in the 1980's/1990's. We chose them over RAW because they gave us a better iNOS yield.

One thing I must stress is that in all research projects, OPTIMISATION is crucial. Researchers generally use a cell line to test an experimental hypothesis......and they will use the cell line that gives the clearest results.

We used J774 in host pathogen interactions because they were available to us in large numbers and they gave the clearest results.

I am unaware of any literature that will compare or contrast the 2 murine cell lines in the way that you are after.

Use both yourself under your lab conditions....... AND THEN CHOOSE!!!!!

Hi, and thanks for your reply.
i am out of idea to choose which cell line should i proceed with for the host-pathogen study since there seems lack of literature search that comparing these two cell lines and pro/cons for using each one of them.
Maybe i shall "borrow" the cell lines and try run for the optimization before i proceed with it.
