bacterial transformation problem - (Dec/08/2011 )

I was doing a bacterial transformation with invitrogen omnimax cells
but i forgot to actually do the heat shock and the SOC medium before

i added the medium but did not do the incubation 37ºC, i went back and did the heat shock and the incubation on ice

its this a problem for the transformation efficiency?

Probably, the conditions for the heat shock are timed based on the heating of a given volume for a specific time. They are also dependent on the concentrations of particular salts in the transformation buffer, which is why you should not exceed 5% of the competent cell volume for addition of DNA.

I donot understand what you trying to ask. Theoretically what ever you did is completely wrong,But if you manage to get a clone you are lucky.
