Anyone use Qiagen pyrosequencer? - (Sep/28/2011 )

Hi all,

We have a problem with the pyrosequencer. As we are looking for methylation, we are always running out of the T nucleotides and Qiagen refuse to sell us single components of the kit, we have to buy a whole new kit.

Does anyone have any solutions?

We want to test the pyrosequencer with different sequencing grade nucleotides but I'm hoping someone else will have a solution to this problem.

try also sequencing the complementary strand? i.e.: biotinylate the opposing strand and then prime off that?
