COL E1 mutations in vector after sequencing - (Sep/13/2011 )

Hi

I've recently cloned two genes into the inducible overexpression vector 'pTRE-TIGHT'.

I've had the whole vector sequenced and it's come back with three point mutations, all residing in 'Fragment containing COL E1 origin of replication'.

I'm wondering if these mutations will have any effect on the overall function of the vector. Or if it's ok to just go on and use?

if you could give more details on the mutations i could give a educated guess if they will effect the vector!
in general any mutation in the origin of replication can effect the plasmid copy number ...maybe your gene is toxic and the mutations will lead to lower plasmid copy numbers so that the burden on the host cell is lower!
