Is there any bacterial vector that can be induced for expression, then turned off with a repressor at later time?

The pBAD vectors made by Invitrogen are regulated by the presence/absence of L-arabinose and glucose. If you want to induce expression, you can pellet the cells and resuspend in media containing L-arabinose and without glucose. When you want to turn off expression, you can pellet the cells and resuspend in media without L-arabinose and with glucose. This system is very tightly controlled, but no system is going to be 100% on and 100% off. You will have to test it out and see if the difference in expression from off to on is enough for what you are doing.
