dephosphorylate sticky-ended vector - is it necessary? (Jul/14/2011 )

Hi,

Currently I'm doing sticky-end ligation. I cut my vector and PCR product (with BamHI overhang) with BamHI.

So, is it necessary to dephosphorylate my vector to prevent self ligation?

Yes ofcourse, you need to dephosphorylate the vector....
