vectors please help - vectors cleavage sites (Jun/01/2011 )

hi,

I was hoping someone couple help me. I have posted 2 links for two vectors.

I was wondering what are the TEV and EK sites? Are they where the tag can be leaved?

I was also wondering what the MC1 and MC2 was?

Hola, both sites TEV and EK sites are recognition sites of proteases one from a tobacco virus and enterokinase and they are designed to give the untagged protein after purification and digestion. TEV protease add a Nt Ser to your protein and nothing for enterokinase. TEV site Glu-Asn-Leu-Tyr-Phe-Gln-Gly/Ser. and Ek site Asp-Asp-Asp-Asp-Lys/. MC1,2 are sites for cloning with restriction enzymes.You have to have all the sequence of both vectors and if the cloning sites aren´t describe introduce the sequences in any REpg. as NENcutter.(by the way you have send only the map of one of the plasmids).
