vectors please help - vectors cleavage sites (Jun/01/2011 )

hi,

I was hoping someone couple help me. I have posted 2 links for two vectors.

I was wondering what are the TEV and EK sites? Are they where the tag can be leaved?

I was also wondering what the MC1 and MC2 was?

Hola, both sites TEV and EK sites are recognition sites of proteases one from a tobacco virus and enterokinase and they are designed to give the untagged protein after purification and digestion. TEV protease add a Nt Ser to your protein and nothing for enterokinase. TEV site Glu-Asn-Leu-Tyr-Phe-Gln-Gly/Ser. and Ek site Asp-Asp-Asp-Asp-Lys/. MC1,2 are sites for cloning with restriction enzymes.You have to have all the sequence of both vectors and if the cloning sites arenīt describe introduce the sequences in any REpg. as NENcutter.(by the way you have send only the map of one of the plasmids).
