Musca domestica gDNA isolation - (May/19/2011 )

Hi people,

I wan to ask if some of you have experience with gDNA extracction from housfly individuals. I use a protocol described in Moreira et.al. (Insect Science, 4:2. 2004) All is going well until I run the gDNA on agarose gel (1%). Togheter with the band over 10kb, I see another band close to 1kb (see pic).

Someone see something like that in gDNA electrophoresis? Partial digesting of gDNA? Supercoiled gDNA? Contamination?

RNA? There can be surprisingly much and large in insects. Add RNAse and see if the band is still there.
