Method to dissolve agarose gel? - (Apr/26/2011 )

Hi, Folks

I have some strains of fungi cultured on 2% Malt Extract agar. I want to dissolve a piece of agar with fungal hyphal in methanol, in order to test some organic compounds in the fungi (ergosterol, similar as cholesterol).

I know it can be done with some commercial kits, but don't know if there is a certain solution that can dissolve the agar.

try heat it up to 60C or to near boiling temperature in water bath?
Since is in agar, can't you possible remove it by using forceps?

NEB sells beta-agarase, an enzyme that dissolves agar.
