Sample preparation for ELISA - (Apr/11/2011 )
Hello
I have to test four proteins in human tear samples based on ELISA method and as you may be aware, I cannot collect enough tear fluid from human eyes. So is there any alternative to increase the initial amount of samples like with a diluent.
I would be very appreciated it if anybody recommend me with which solution and how can dilute human tear sample for ELISA experiments?

you can always dilute samples using a sample diluent, however the signal depends on the amount of the tested protein in your samples.

Which is the best dilution?
can I dilut with PBS?
A quick search on google indicates various buffers used in the litterature. I would probably stay with PBS...hope your assay has enough sensitivity. You can always increase the incubation time or incorporate polymers to increase sensitivity.
