if I'm doing cell cycle and expecting high proliferation rate,
where I need to expect the cells to be s phase or g2

Please use a positive and a negative control, for example serum starved cells (G1 enrichment) and Nocodazole (G2/M enrichment). This will tell you where your G1 and your G2/M signal is. The signal between those two peaks is S phase.

I know the distribution between the phases, my quesion is if for example I'm looking at proliferating cells
and I'm getting more in G2/m is this comparable or I should expect them to be in S phase

I don't completely understand your question... Comparable to what?
You will have a distribution, with a certain percentage in G1, S and G2. If you have a "treated" cell sample, those percentages will change, for example, you have 25% in G2/M phase instead of 15%. So your treatment (gene) induces proliferation compared to control untreated cells. You usually compareG1and/or G2/M phases, S phase is rather transient without cell cycle checkpoints (Errr, was it?).
