Filter for viral transfection - filter 0.22 or 0.45 (Apr/04/2011 )

I do viral transfection (retro) made with Plat-E cells,
and I normally use 0.22um filter to remove cell debris from supernatant
but I just found some papers using 0.45um filter instead.

which is better? or no change??

Let me know your opinion or routine.

Thanks in advance

For the virus it shouldn't matter, both filter sizes are much bigger than a virus. 0.45 will get rid of most bacteria I think. A 0.22 um filter will get rid of all but mycoplasma.

In general people do not recomend the 0.22 filter (they specifically mention it in most protocols)because you reduce the titer of the virus big time.
I always use 0.45 filters (but I never checked what happens with the 0.22 filter)

thanks everybody
I will switch to 0.45

Thanks

Yep. Me too. I always use 0.45 syringe filter.
