cell culture media - glutamine! - (Mar/17/2011 )

I use Ham's F-12 media for CHO cell culture

My media composition suggests that i use media with 0.2mM glutamine along with other media constituents like 10% FBS 1% P/S

But the media (500ml) we buy always contains 1mM glutamine. How would I prepare media with 0.2mM glutamine considering that the media already contains glutamine

1) Will something BAD happen if i use media with 1mM glutamine?

2) How would i prepare media with all my constituents and still contain 0.2mM glutamine?

You will not be able to prepare medium with 0.2 mM glutamine given that it already contains 1 mM. You could however buy glutamine free medium and prepare from there.

I don't know if excess glutamine will have any effect on the cells.

FYI - Glutamine does degrade quite quickly in medium.

You wont be able to lower the concentration of the glutamine in the media as it has already been added. Also you do not want to dilute the media as you will dilute all the other components which are probably at a level that accomodates cell growth. There are plenty of mediums (off the shelf)
that you could use.

I would try the glutamine at 1mM as it is pretty low anyway for CHO cells (if that is what your using).

Glutamine becomes a problem when a high ammonium concentration in the culture becomes an issue. As ammonium is a by product of glutamine. However at 1 mM this shouldnt be an issue. Some cells do not require glutamine and may only require it at the beggining as they will start to produce there own which is a desirable characteristic
