New to Gene cloning - Plasmid vector preparation (Mar/10/2011 )

Hello all,

I am a cell culturist and completely new to Molecular biology stuff. All I do is get the plasmid vector / DNA / virus from another group, do transfections, grow the cells, do selection, analyze the cells for expression using various cell based assays, FACS, clone and develop stable cell lines expressing recombinant protein.
But I have never done the plasmid vector generation and would like to do some of that part in my job. I don't want to ask my boss looking like a fool. I know this is basic to molecular biology. Can you suggest a broad methodology / web links on how I can get started? I know there is lot of info on the web - which is confusing to me.
I am not completely naive to the subject - cause I've done DNA purification, restriction digest, running the digests and doing southern blotting in my old job.

you can go for the Qiagen Bench Guide ...very useful for your purpose!
