Tell me how you would perform this split - (Feb/11/2011 )

Starting with a T75, perform a 1:7 split into 15, T25s.

Assume T25 plating volume is 5mL.

You can't do it. The 1:7 is on the original one, so at most you can get 7 flasks.

But you're going into a vessel that's a third of the size as the original one.

Still, 1:7 is 1:7, if you need to split it 1:15, then do that... split ratios are independent of area, they are based on taking a proportion and it doesn't matter what area you are seeding into.

Now, if you had said that you wanted t-25 flasks seeded so that they are 1/7th confluent, that is a different story. A t-25 will give you about 3 x106 cells when confluent, so divide that by 7 and you have your answer.
