why the cells transformed with linearized plasmid cannot grow? - (Jan/09/2011 )

Hi, everyone,

i would like to know that will the linearized plasmid still can grow after introduction into competent cells?

-chi-

No, since linear plasmid DNA can not be replicated.

thx.

is that mean the cell can take up the linearized plasmid but the plasmid cannot replicate inside cell?
and also, why the plasmid cannot replicate?

-chi-

linear DNA can be taken up by cells, that is not the problem ...the problem is that replication is dependent on a circular molecule, and a double strand break can not ligated by E. coli.
