reconstitute problems - antibody from R&D systems (Dec/22/2010 )
My friend bought an antibody(solid) from R&D systems .But she was confused when she tried to RECONSTITUTE it,which is called TGF beta 1.Purified recombinant human TGFβ1 is an extremely hydrophobic protein that adheres strongly to surfaces. To ensure recovery, reconstitute at 20 μg/mL in sterile 4 mM HCl containing 1 mg/mL human or bovine serum albumin,the reference from R&D says.
Here's the problem,how to reconstitute an antibody?Should she use a PBS or simply sterile water to reconstitute?Does anybody have a specific protocol?Thanks in advance!

Hola, the supplier says the correct way to reconstitute, first prepare the 4mM HCL. Take 0,5-1ml and dissolve 0.50r 1mg of BSA. filter in sterility the solution and add to the liophilized powder the amount of solution necessary to have 20ug/ml. 0.5 ml to 10ug of powder or 2.5ml for 50 ug. the amount of powder will be in the vial isnīt it?. Thanks for helping me.Some people may fed up with this stupid question.However,as far as I am concerned, success depends on details.One more question, does liophilized here mean lyophilized?

Is your friend trying to reconstitute the Ab or the TGFβ1? Abs are usually reconstituted in dH20 to a concentration that is recommended by the manufacturer so as to get a salt concentration that is optimal for the Ab.
