Antibody gene cloning - (Dec/09/2010 )

Dear forum members,

could anyone tell me how to amplify the gene sequence of an antibody having the mRNA (or cDNA)? What I mean is if there is any consensus sequence in the V-gene and C-gene despite the huge variability of these genes.
Thanks a lot in advance.

You can use degenerated primers or a combination of primers that amplify most encountered sequences. However, these are species dependent - different for mice and humans. I know a good set of primers for mice, humans I have not done before, but I believe it is much the same.

Thanks once more! Could you pass me any reference/paper? I am working with mice antibodies too.

Thanks again for the papers!
