IC50 - (Dec/01/2010 )

Hey guys

Could one of you geniuses help me with this. Ive been given a table with inhibitor drug concentrations and another two column with enzyme activity in the presence of either drug. I need to calculate IC-50 values. Ive figured I need to draw a graph. What do I plot on the graphs?! and also..what can I comment or infer after seeing the values?

pay attention for IC50 the graph is sigmoidal.
i.e : the X-axis is log ; X : conc.

Until now, I have a vague understanding in determining ic50. Different software gave different values. What is the minimum point for accurate IC50 determination. In my cases, the inhibitory effect at low concentration will vary between replicates and not at high concentration. When all points were selected, the varied value at lower concentration will make large difference of linear equation between replicates. When I chose only three points, I got good result with small SD. I dont know this is acceptable as I cant find any articles specifically discuss about ic50 determination

you may want to ensure that your pipettes are calibrated and reproducible. major variation of replicates is not acceptable.
