Glycine linker - (Oct/05/2010 )

Hello all,
I am preparing a expression vector and I would like to get your advice on glycine linker. I have a gene of interest and to over express and I have to insert a RFP gene somewhere middle of this gene to see the localization of our gene of interest. I am totally unsure about the glycine linker which I am supposed to add before and end of the RFP gene. Please advise me the points which I have to keep in mind regarding glycine linker, like number of glycine residues and what are the criteria for the selecting the numbers etc. Thanks in advance.

Usually linkers are a mix of glycine and serine residues, often the sequence GGGGSGGGGSGGGGS on the theory that the serines provide a bit of hydrophilic nature to the linker.
