what is the optimal temperature for my restriction enzyme - Urgent plea (Sep/14/2010 )
What is the optimal temperature to use for my RE Xba1 ??
Thanks, I haven't use this before yet..
Hola, Check any catalogue and it shows all the specifications of each RE. I have seen a Takara poster and it says 37ºC with media salt buffer and BSA. Good luck
I can give u the optimum temp for XbaI but if u want to check for other enzymes in future, thatswhat i am suggesting u to check in the neb site.
Temp is 37 (http://www.neb.com/nebecomm/products/productR0145.asp) Check the link.
And one more thing about XbaI is the flanking bases of the restriction site (dam methylation). I had a bitter experience, after that i changed the flanking and worked out well.
All the best.
