Removal of Tightly Bound Nucleic Acids - (Aug/07/2010 )

Hello,


I am having some trouble purifying a DNA binding protein. It seems to be bound by some very strongly interacting nucleic acids, from the cell lysate, as suggested by the very high absorption at 260 nm on the UV.
Could you suggest any protocol(s) which will allow for the removal of such nucleic acids?

Thanks for the suggestion, already tried adding some, did not work. Next time I'll add a bit more...

perhaps you should try to use an exonuclease (the "normal" DNAse is an endonuclease).
