why my protein precipitates? - (Jul/28/2010 )

1) i am trying to crystallize my protein and for that i lyophilized my protein sample. After obtaining in powder form, when i tried to redissolve it in water, it precipitated. Why?

2) I have another batch's protein where i think lyophilization was a bit incomplete in the sense that it still is flaky, this one behaves very transparent upon redissolving. Why?

Can somebody throw a light?

Hi,

Maybe during the lyophilization process your protein changed its structure, you can try redisolved in DMSO and water.

think "degree of hydration". just like with nucleic acids, some proteins don't like to be dried completely.
