Stimulation of transcription - (Jul/26/2010 )

Hi Everyone
I am very new to the cell culture scene so i was wondering if anyone can help me?
I have a T cell line and i want to stimulate transcription of my gene of interest. How do i find out what would stimulate transcription of my gene?
Also can you buy transcription factors? if so where from?

First you have to know where is your gene involved, which pathway...

It could be PMA,Ionomycin,LPS...

The gene is involved in intracellular transport along microtubules. The only pathways i can find are cytoskeleton remodeling and cytoplasmic microtubules.
However i have found some transcription factor binding sites in the promoter pol2, NFkappaB etc.

well, so you can use something that activates NfKB, and see what happens

Where can i buy purified LPS?
