Storage of Fixed Cells for Immunofluorescence staining - (Jun/30/2010 )
Dear all,
I want to do a time course experiment with a tumor cell line over 10 days. I have to induce every plate of cells at one day and then harvest every day over ten days. I cannot do it vice versa. This is a problem, as I will simultaneously need to make samples for IF (on coverslips in the 10 cm plates). This would mean I have to do staining every day, which is 4 hours for 4 slides or so... I would prefer to stain on one day and store the coverslips somehow until then. Also, I would have the same staining conditions, then.
Do you know a method to keep fixed cover slips for several days until staining?

Just store them at 4 degree in PBS. I do this all the time and you can hold coverslips for weeks.
