What is the solution when tween80 or DMSO is not enough to dissolve plant extract?
Hi,
I'm stuck on this thought, can you please help me!?
I'm study the effect of some plant extracts (by organic solvent : hexan. chloroform, EA and methanol) on nematode larvae.... I have to use just 3% of DMSO or Tween80 to dissolve 5 grams of extract to prepare 100ml of 50mg/ml extract as stock by dissolving 5g of extract in 3ml of DMSO or Tween80 and add 97ml distilled water and then I'll use this concentrations 50, 25, 12.5, 6.3 and 3.2 but the problem is 3ml is not enough to dissolve 5g of extract and If I add more DMSO OR Tween it'll be toxic for the larvae?
So, how to solve this problem

Can you just dissolve the extract at the final concentration directly in the culture medium for the nematodes?

Can you combined both the DMSO and Tween for the 1st preparation?
Can a different solvent be used...DMF, cyclohexane, methanol?
If the extract is not expensive you could weigh small amounts and check soulbility with a number of organic solvents...
Is the extract(s) or similar compounds listed in Merck Index which has solubility info?
If you are purchase the extracts will the supplier have info helpful to you?

I prepared the extract by myself using 4 organic solvent (hexan, Chloroform, EA and Methanol) and after drying of extract I have to prepare a stock using small amount of DMSO or Tween to improve the solubility of extract in water and I cant use any organic solvent that you mentioned because they affect the larvae and if I used them to my extract and added water and then evaporated the organic solvent I will get clusters of extract.

Well can you then try another non-toxic solvents such as acetone that help solving lipophilic and hydrophilic substances. It will evaporate too later.
another possibility would be to reduce the amount of extract. 50mg/mL seems quite a lot (?). To use more DMSO or Tween you can give a try. As you need anyway a control with solvent and auxiliary substances, you will see if those substances have an influence on mortality, i.e. if they increase it.
Cyclodextrin is another option and if that doesn’t work I usually look to suspend plant compounds in solution with binding proteins, eg BSA or pure serum.
I hope you have enough sample to keep trying.

Well have to arrange a supply utilizing little allowance of DMSO or Tween to advance the solubility of extract in water and I cant use any organic solvent that you cited because they sway the larvae and if I utilized them to my extract and supplemented water and then evaporated the organic solvent I will get clusters of extract.
