About gene orientation - (Jun/07/2010 )

Hi, i just wanna know what is the effect of gene orientation in a recombinant construct on protein synthesis. Consider following two cases.

TER1-GOI-PRO1---PRO2-GOI2-TER2

PRO1-GOI1-TER1---PRO2-GOI2-TER2

in case 1 expression will proceed in two directions and therefore read-through transcription from one transcript to the other will be prevented.

in case 2 expression goes in one direction and although there is a terminator after GOI1 there will be read-through transcription from promoter1.

Hi,
I am just wondering if there is a terminator in between, then how a read through transcript is possible originating from promoter 1.
Another question is what if i use same promoter for both transcripts in case 1. Will there be any steric hinderance?
