question about plasmid extraction - (Jun/04/2010 )
Hi all
After cloning(by fermentas pTZ57R/T cloning vector) and plasmid extraction, products detected on 1% agarose gel...
I saw three bands on gel.
Is it common, or no?

don't worry.
Vectors have several conformations:
The supercoiled form is the one that runs quite fast.
Then there is the nicked circle form which runs slowly.
And the last typical one is the linear form where the circular strand is broken. It runs above the super-coiled form.
Sometimes you may also see dimeric structures.
And also methylation has effect on the conformation.
