anion exchange and buffer - (May/26/2010 )
I have a biomolecule that im trying to purify using anion exchange (the biomolecule is negative charge). The problem is is that the biomolecule is in phosphate buffer at ph 7.3. Is it alright to put this on an anion exchange column? My thoughts were that it wouldnt bind since the phosphate is negative charge as well. Is this correct? Thanks

you should have no problem. the phosphate (or other buffering component) will not interfere with the binding of your protein.
