dye for practice brain injection - (May/19/2010 )

Hi,

I'm going to be injecting toxin into mouse brains using a stereotaxic device. But before I inject my precious experimental mice, I want to make sure that I have my coordinates right. So I want to inject a dye into the brain for my practice run, fix the animals, section the brains and see if I hit my targert correctly. Does anyone have any suggestion as to which dye to use? Thanks.

You could try Evan's blue, which is what we used for mouse foot pad injections.

Pontamine Sky Blue is another good option. Fast Green could work as well.

I've heard of Evans blue and fast green, but not pontamine sky blue. Any suggestion on concentration?

India Ink at a dilution of 1:100 is another option. I've used it for practice injections, but in my case I did not fix tissue after injection, simply harvested the brain, made a coronal cut with a blade at site of injection and checked under dissection scope.

any dye would work, even trypan blue.
