Two peaks measured by DCFH-DA - (May/16/2010 )

Hi,
I have tried using DCFH-DA in assessing the ROS generation in my cells (Caco-2). It is supposed that with my sample added, there should be less ROS generated. However, after I did the flow cytometry, I found out that there were two peaks in the histogram of the control and only one peak was shifted (but not the other) in the cells with sample added.
Does anyone know the reason behind?
Is it the interactions between my cells and samples are not good enough so that only a portion of cells can respond to my sample?
And can I conclude that my sample is effective in decreasing the ROS generation in Caco-2 based on this result? Or shifts in both peaks are necessary in getting a conclusion?
Please help me.
