tagged mouse - (May/15/2010 )

i need to make a mouse that has a tag on my desired protein. The application will be for immuno-precipitation and immuno- fluorescence (FISH) mostly. Does anyone have a knowledge about this subject, like which tag (FLAG, HA, MYC, GFP), if it better on the N or C terminus, shold i fuse with it a linker of glycine for example or fuse straight to my protein?

I have not made transgenic animals myself, but I can direct you to a few papers.

These are links to some papers by Barmada and Ivanova that describe prion-GFP GMO mice. They discuss how they were made, how they determined the placement of the GFP tag, etc. That should get you started.
