Could a 30aa peptides(3×flag+6×his) be expressed by BL21 and be successfu - What is the minimum of the length of expressed protein? (Apr/22/2010 )

Could a 30aa peptides(3×flag+6×his) be expressed by BL21 and be successfully purified by Ni-beads?
I want to make some homemade 3×flag peptide for elution of protein from M2 beads.But I suspect if it could be readily expressed and purified and detected as it is so tiny.
If you don't think so,do you think some 10(or more)×flag +his tag could work?

expression of small peptides should be no problem for E. coli ...it could be that small mRNAs are unstable ...that coudl be a problem ...but in general it shoudl be possible.

What we experienced is that the cells are heavily affected by induction of an empty pET30a vector where only tags are expressed. But they produce them. Thats is all i can say.

Purification? ...i have no experience on the purification of such small peptides but i would suggest that it is also possible.
