Generating stable cell line - (Apr/09/2010 )
Hi all,
I am a bit confuse of the definition of stable transfected cell lines. Is the vector being integrated into the host genome or the vector is still in the protoplasm of the host? I thought that only the retroviral vector will be in integrated into the host genome.

stable cell lines are the one in which the vector containing your gene of interest will integrate into the genome.the vector has sequences that help in the integration. for this, mostly linearized vector is transfected.

what will be the vector of choice with the preferential sequence? or will any linearised vector do the job? im planning to generate a stable cell line but dont know if i should purchase a specific vector for the job or just use any in-house vector...thanks for answerin!
