CO2 production by glutamax - (Mar/24/2010 )
In our lab we would like to allow cells to shake a couple of days in an incubator that is not connected to a CO2 supplier.
However, we need to obtain 10% CO2. We thought we could do this by addition of glutamine-glutamax to the culture medium.
Unfortunately, I've not been able to find a lot of relevant information about this method of CO2 production.
Does anyone have experience with this? And if so, do you have any idea how much glutamine-glutamax has to be added to the
culture medium in order to obtain 10% CO2 production?

Use a media formulation that does not require the CO2-bicarbonate system for pH balance. You could do this using HEPES for example or could use a bicarbonate free media such as L-15 (Leibovitz). This media was designed specifically for this purpose.
Glutamax is just a more stable form of l-glutamine, which is an essential amino acid. It will not generate CO2 in solution, nor will it balance pH.
