Moisture in Agar plates! - (Sep/24/2009 )
Yesterday I was pouring a ton of plates (100 15cm plates), and now there is a ton of moisture on the agar itself. What is the best way to get rid of it now? Vent them upright and let them dry? They have Kanamycin on it, but I don't want the plates to get contaminated.
Hi,
Did you let the plate to cold down to room temperature in the laminar before closing their lids prior to storage in chiller?

What was the temperature of the media when you poured the plates? It sounds like there was a lot of condensation on the lids, and when it cooled, the drops fell on the agar. There's no worry - since the condensation came from the media, and they have Kanamycin in them, chances are slim that they will get contaminated. You can invert the plates and allow them to sit overnight; however, you don't want them too dry - the kanamycin will degrade without moisture.
Always wait for the medium to be cool enough to be grabbed in hand conveniently and after pouring the plates (with the bunsen on) dnt put lid immediately wait for them to be dry .......(place the lid halfway over it)........when they solidify and they still contain moisture u can use incubator to dry them (place the lid beneath and the plate inverted half way over it)....it will take 15 minutes to become dry.....
