Theoretical question about retroviruses - (Sep/11/2009 )

Hi there,

I was wondering if one of you guys could answer me a question about the moleculular behaviour of retroviruses. I am trying to subclone a CAG promoter into a propietary retroviral vector and, since the promoter carries a large synthetic intron, I am afraid that during the production of retroviral genomic transcripts the intron is lost and the function of the aforementioned promoter is compromised.

I also have been reading papers that describe the production and titulation of lentiviral vectors that carry the CAG and, besides a slight reduction in the viral titer versus the parent vector, they seem to work fine...

¿What do you think about that?
