increase the amount of cDNA - (Sep/08/2009 )

Hi!

Is it possible to produce more cDNA from cDNAs samples? I cannot reverse transcribe my RNA samples because they are over...

Thxs!

Isn't that what PCR is for? :lol:

BD makes a cDNA amplification kit.

Are you doing RACE?

You can, but you have to careful;
- If you re-input too much cDNA, it will inhibit your subsequent PCR
- cDNA contamination of the general working area can become a serious issue
- You may start to get non-specific products driven by the sequence homology and high concentration of the cDNA template (increasing the tm may help here).
- I would not rely on the second round of PCR for any quantitation.
