How long do you warm up media? - (Sep/02/2009 )

Any damage could be done to media if I warm it up too long?

Say....like half of a day before use?

I just forgot that I put it in 37'C water bath in the morning..

It was a one of those crazy days that i was mentally off..

Then 6 o'clock in the evening, same day, I just found a bottle of media, and was like "Hmm...that one seems familiar.........uh?......argg! it is mine!!!"

Would it be okay after extra hours of warming?

If not, why??

if it is plain media I would go with it.

I did forget one bottle over night at 37C. It was perfectly fine.

what media do you mean and for what? And was whatever you wanted to put in the media allready in it?

when I grow cells I put the media at 37°C the evening before I use it, and sometimes even 1 day before that.
The media wont go bad... I so no reason why to worry.

Cell culture medium containing L-glutamine should only be warmed for short periods of time as the L-glut is unstable. Long periods in the waterbath will cause it to degrade.

i never warm up the media in a water bath. I just take it out in the morning and bring it up to room temperature. and I never had any problems with both established lines and primary cells.

Ya. I would do the same if don't need the media immediately.
Otherwise I'll warm it in waterbath for 30 min if I am in hurry.

I warm mine up in the waterbath for 30 min, too. I start the hood, put the medium in the waterbath, and have a cup of coffee. Then I'm ready to go. :) However, long periods of incubation at 37 C will also cause amphotercin to degrade as well as the L-glut.

Amphoterecin???? you don't want that in your culture, antibiotics and especially antimycotics are bad - they can really screw up your culture and mask low level infections which will cause spurious results.
