Multiplex Reverse Transcription? - Maximizing limited RNA samples (Mar/09/2006 )
I am doing qPCR but have very limited RNA from our primary cultured neurons. I have tried doing the reverse transcription with random hexamers but have not had any luck in detecting my gene of interest this way. I think it is due to its very low expression level. I can detect it with a specific for RT. However, since I have limited sample, doing it all my specific primers is a big waste of material for me. Is it possible to do "multiplex RT"? For example, include the specific primer in the reaction with the random hexamers or do the RT with a number of specific primers?
