Collagen synthesis assay - (Oct/25/2005 )
Does anyone have any experience doing assays for new collagen synthesis for cells imbedded in collagen matrix. The collagen matrix is bovine so one option is to perform an ELISA for human collagen (the cells are human). Second approach is tritiated proline incorporation. I am looking for specifics on how you treat the collagen matrix prior to your assay. I have seen the Chondrex collagen assay protocol and that uses combined pepsin and elastase digestion to create monomeric collagen that is tested by ELISA. How about with the tritiated proline? Thanks for your help.
