Standard Protocols

Haematoxylin and Eosin Staining

 SOLUTIONS

(i) Mayers Haematoxylin (filtered)

(ii) Eosin

Eosin Yellowish 1.0g

Distilled Water 100ml

(iii) Acid Alcohol 1%

70% Alcohol 99ml

conc. Hydrochloric Acid 1ml

 METHOD

  1. Take sections to water.
  2. Place sections in haematoxylin for 5 minutes.
  3. Wash in tap water.
  4. 'Blue' sections in tap water.
  5. Place sections in 1% acid alcohol for a few seconds.
  6. Wash in tap water.
  7. Place sections in eosin for 5 minutes.
  8. Wash in tap water.
  9. Dehydrate, clear .
  10. Mount sections in Immumount or DPX

 

RESULTS

Nuclei Blue-black

Cytoplasm Varying shades of pink

Muscle fibres Deep pinky red.

Fibrin Deep pink

Red blood cells Orange/red
