primer optimization - (May/22/2005 )

When i do the primer optimization should i use my sample or should i use other samples which i don't need with my target to find the optimal primer concentrations? If i use my sample for this optimization will not i waste my sample? How do you do?

Hi indoubt,

Usually it depends on how difficult it is to get your template and how precious it is to you. If sample is limited I would try to optimise on something else that does have my gene of interest.
