Storing of miniprep or maxiprep plasmids - (Mar/23/2005 )
In which solutions and where should i store my miniprep or maxiprep plasmids? How long can i store them in this place before they get damaged? When i use them can i thaw them with my hands and will they get damaged if they are subjected to many freeze and thaw circles?
Thanks.
hi
for years at -80 in solution or dried
for month at -20° in solution or dried
but if your prep is very clean you can store for three years at -20°C in solution
solution is generally TE buffer (allows better resuspension than ddH2O specially in cases of maxipreps)

hi
thaw them on ice it will reduces damages. but i make a reference that allow me to re transform bacterias if i xant a ne prep and several aliquots if i plan many thaw for my job
it's essential to store a reference tube.

I thought we should only store the vector in bacteria and make a new culture from this to get new vectors. Any ideas?
HI

I think it's better to save both. but bacterias are a quicker way to get back preps...

In summary; after you make the mini or maxi prep you store the vector as aliquots in TE or water and thaw them when you need them? What volume do you make you vector aliquots ?
Should i always keep my vector on ice when i work?
Thanks again.
I store my DNA in buffer EB in -20C. As long as your prep is clean, it should be fairly stable.
I thaw them on the bench and leave them there until I need them.
I've once sent some DNA for sequencing. It was just a styrofoam drop-off box, it got left there for 5 days. Came out ok.
