Troublesome Cre-loxP...Anyone experienced can help me?～ - (Nov/19/2008 )

Hi~
Recently I am using Cre-loxp to investigate a certain transcription factor in primary neuron culture.
We are using AAV-Cre to infect the primary cultures. However, it seems it take really a long time before the Cre recombinase to take effect. I tried for one week, but it still did not work...
Has anyone used AAV-Cre in primary neuron cultures before and could you share me some experience, for example, how long does it take before cre works? I wonder whethet it is the problem of my virus...

for the factors we are looking at , it take 3 days to see clear reduction of the protein.
We usually look for recombination by PCR and do Immunostaining as well as western blot to verify the protein's level.
